Genetics of Asthma: Insights From Genome Wide Association Studies

Abstract

Asthma is a heterogeneous respiratory disease that can be influenced by genetic and environmental factors leading to disease susceptibility and progression. Genome-wide association studies (GWAS) of asthma have yielded replicated findings and identified novel risk alleles and loci demonstrating the polygenic nature of disease. These studies identified roles for genes related to Type 2 inflammation, airway structural changes and altered epithelial cell barrier. In the current review we summarize these findings and discuss more recent GWAS studies of subgroups of asthma which have provided additional insight into disease pathogenesis. These genetic findings provide new understanding and therapeutic opportunities for asthma.
